Recent research has identified specialized core promoter elements that cater to specific gene families. The TCT motif is a prime example, found almost exclusively in the promoters of ribosomal protein genes.

Unlike most protein-coding genes, those containing a TCT motif do not utilize TBP/TFIID for initiation; instead, they are recognized by the TBP-related factor 2 (TRF2), creating a distinct regulatory pathway for the cell's translation apparatus.

Similarly, the X core promoter elements (XCPE1 and XCPE2) have been identified in TATA-less promoters, where they facilitate TFIID-independent transcription initiation.

These findings suggest that the core promoter is not a passive component but an active filter that determines which enhancers and activators can successfully stimulate transcription.

Unlike TBP which heavily distorts the DNA with a 90° kink, TRF2 binding to the pyrimidine-rich TCT motif maintains a relatively linear, straight double helix, interacting specifically with the unique geometry of the ribosomal initiator.
